Workshop I: 3-MCPD and MOSH/MOAH Analysis - Learn about the details of the automated workflows for routine analysis

About the workshop

3-MCPD (3-monochloropropane-1,2-diol), glycidol and the MOSH/MOAH mineral oil substances are regulated contaminants in many foodstuffs. The manual sample preparation is complex with many steps and prone to operator bias making it difficult to reproduce and compare results. The control of 3-MCPD and glycidol in oil raffination processes, in particular for palm oil products, requires fast and reproducible analysis methods. Food contaminations by mineral oil hydrocarbons (MOH) can occur during food processing and from food contact materials. In both cases, 3-MCPD and MOH, serious impact on consumer health due carcinogenicity is reported and led to regulations with maximum residue concentrations that makes reproducible trace analysis of these compounds on global level indispensable. This workshop presents the fully automated sample preparation methods and analysis according to the international regulations and standards have been developed and are in use in governmental, industrial and private laboratories globally.

The automated analysis of 3-MCPD, also 2-MCPD and glycidol in edible oils uses the AxelSemrau CHRONECT Workstation MCPD with the automated hydrolysis of the fatty acid esters, then derivatization using phenylboronic acid. Analysis is done online by GC-MS or GC-MS/MS. All international ISO and AOCS standard methods have been implemented as automated workflows.

The automated MOSH/MOAH analysis in food requires the separation of the saturated hydrocarbons from the cancerogenic aromatic compounds which can only achieved by a normal phase liquid chromatography. The complete automated workflow using the CHRONECT Workstation MOSH/MOAH including the removal of biogenic interferences from alkanes (AlOx clean-up) and from natural olefins by a proprietary epoxidation step to avoid the overestimation of the critical MOAH content, is presented.

About the speaker

Hans-Joachim Huebschmann

Hans-Joachim Huebschmann graduated as Certified Food Chemist from the Technical University in Berlin, Germany, working on residue analysis of anabolic agents and pesticides in foods.

Hans started his career in diagnostics, medical analytical instruments and application development working for a number of German companies in chromatography, mass spectrometry and in biotechnology. In 2002 he joined Thermo Fisher Scientific in Bremen, Germany, and held several international positions covering inorganic and isotope ratio mass spectrometry, high resolution GC-MS and GC-MS/MS, specializing in food safety, forensic and dioxin analysis. In 2015 Hans established the independent agency HANS Analytical Solutions active as independent consultant for companies in Europe and Asia for the development of automated analytical sample preparation workflows. In his current international capacity Hans is located in Europe and Osaka, Japan.

Hans is the author of the popular Wiley "Handbook of GC-MS", available internationally already in is 3rd edition in 2015, 776 pages

Workshop II: Compliant modernization of HPLC gradient elution methods in view of USP 621 revision

About the tutorial

In this workshop, we will discuss approaches for development, optimization and transfer of legacy and modern HPLC methods, utilizing gradient elution. This will also include the method modernization or the transfer from HPLC to UHPLC. Discussion topics and data on how to more easily move to core-shell particle technology, how to modify gradient methods effectively, and how to work within the framework of evolving rules for method transfer such as USP 621 or the respective chapters in EP or JP will be addressed. The latter aspect is very topical, as these monograph chapters had all undergone a recent revision to fully address gradient-elution-based method modernization and adaptations. Starting from the relevant theoretical background, most importantly Snyder's gradient volume principle, we will also discuss the use of effective tools to facilitate method transfer across HPLC platforms. The principles will be illustrated with simulated chromatograms using method development software, but more importantly, with experimental data from relevant pharma applications.

About the speaker

Paul Gamache

Paul Gamache is an analytical chemist who joined Thermo Fisher Scientific in 2011 through acquisition of Dionex Corporation. He is a Director of Research and Development responsible for development of analytical technologies with primary focus on charged aerosol detection (CAD) and electrochemical detection (ECD) for LC. Prior to joining Dionex in 2009, Paul helped to lead development of first and second generation Corona CAD products. Paul is editor and contributing author to the book "Charged Aerosol Detection for Liquid Chromatography and Related Separation Techniques" published in 2017 by John Wiley & Sons, Inc. and is a member of the ASTM International committee on Nano-enabled Medical Products.
